Depazide-GM1 (Dapagliflozin 10 mg + Glimepiride 1 mg + Metformin HCl 500 mg) Tablet is a scientifically designed triple-combination therapy for the effective management of type 2 diabetes mellitus. It combines dapagliflozin, glimepiride, and metformin HCl—three well-established antidiabetic agents with complementary mechanisms—to deliver comprehensive glycemic control. This combination helps target multiple pathophysiological defects, improving blood glucose levels while supporting cardiovascular and metabolic health. 

It combines three actions: removing excess glucose via urine (dapagliflozin), increasing insulin production (Glimepiride), and improving insulin sensitivity (metformin). Dapagliflozin lowers blood sugar by causing the kidneys to get rid of more glucose in the urine. Glimepiride lowers blood sugar by causing the pancreas to produce insulin (a natural substance that is needed to break down sugar in the body) and helping the body use insulin efficiently. Metformin helps to control the amount of glucose (sugar) in your blood. It decreases the amount of glucose you absorb from your food and the amount of glucose made by your liver. Metformin also increases your body's response to insulin, a natural substance that controls the amount of glucose in the blood. 

How does Depazide-GM1 (Dapagliflozin 10 mg + Glimepiride 1 mg + Metformin HCl 500 mg) work?

Dapagliflozin 10 mg (SGLT2 Inhibitor) 

It is a selective inhibitor of sodium-glucose cotransporter 2 (SGLT2) in the proximal tubule of the kidney.It blocks the reabsorption of glucose from the urine back into the bloodstream. This promotes the excretion of excess glucose through urine (glycosuria).

Reduces blood sugar independent of insulin, promotes weight loss, and provides cardiovascular/renal protection. 

2. Glimepiride 1 mg (Sulfonylurea)

It is an insulin secretagogue that acts as a third-generation sulfonylurea.

It stimulates pancreatic \(\beta \)-cells to release insulin by blocking ATP-sensitive potassium channels (\(K_{ATP}\) channels).

Increases insulin levels to lower blood glucose, particularly after meals. 

3. Metformin HCl 500 mg (Biguanide) 

It acts as an insulin sensitizer, primarily targeting the liver and intestines.

Liver: Reduces hepatic glucose production (gluconeogenesis).

Intestine: Slows down intestinal glucose absorption.

Peripheral Tissue: Increases peripheral glucose uptake and utilization.

Decreases insulin resistance and lowers fasting/postprandial blood sugar without causing hypoglycemia.

Dosage & Administration

To be taken as directed by the physician.

 Usually administered once daily with meals to reduce gastrointestinal discomfort and risk of hypoglycemia.
