Depazide-10M

Depazide-10M (Dapagliflozin 10 mg + Metformin HCl 500 mg) Tablet is a clinically effective fixed-dose combination of Dapagliflozin 10 mg and Metformin HCl 500 mg, designed for advanced management of Type 2 Diabetes Mellitus. This combination therapy combines the insulin-independent glucose-lowering effect of an SGLT2 inhibitor with the proven efficacy of metformin to improve glycemic control, reduce insulin resistance, and support long-term metabolic health. This reduces the chances of serious complications of diabetes and also helps prevent heart disease. The dual-action therapy is used together with proper diet and exercise to treat type 2 diabetes. It is also used to lower the risk of hospitalization for heart failure in patients with type 2 diabetes and cardiovascular disease or multiple cardiovascular risk factors. Dapagliflozin and Metformin. Dapagliflozin works by removing excess sugar (glucose) from your body through urine. Metformin works by lowering sugar production in the liver, delaying glucose absorption from the intestines, and increasing the body's sensitivity to insulin. Together, they provide better control of blood sugar. 

How does Depazide-10M (Dapagliflozin 10 mg + Metformin HCl 500 mg) work?

Dapagliflozin (10 mg)—SGLT2 Inhibitor:

Inhibits sodium-glucose cotransporter 2 (SGLT2) in the proximal renal tubules.

Reduces renal glucose reabsorption, lowering the renal threshold for glucose, and increases urinary glucose excretion (glucosuria).

Promotes lower blood glucose, weight loss, and reduced systolic blood pressure.

Metformin HCl (500 mg) - Biguanide:

Decreases hepatic glucose production (gluconeogenesis), decreases intestinal absorption of glucose, and improves insulin sensitivity by increasing peripheral glucose uptake and utilization.

Lowers fasting and postprandial blood glucose without causing hypoglycemia. 

Combined Action: The combination provides a dual mechanism of action—targeting both the kidney and liver/muscles—which results in improved glycemic control (HbA1c reduction) compared to individual agents, often with added benefits of weight loss. The 10mg/500mg dose is often used in a fixed-dose combination. 

Clinical Benefits 

The combination effectively lowers fasting and postprandial (after-meal) blood sugar, with studies showing significant improvements in \(HbA_{1c}\) (often a reduction of \(\geq -1.1\%\) to \(-1.6\%\)).

Dapagliflozin 10 mg lowers the risk of cardiovascular death and hospitalization for heart failure in patients with type 2 diabetes who have established cardiovascular disease or multiple risk factors.

The combination helps slow the progression of kidney disease (nephropathy) in diabetic patients, reducing the risk of end-stage kidney disease and worsening of chronic kidney disease.

Unlike many diabetes medications that cause weight gain, this combination promotes weight loss (an average reduction of \(1.4\)–\(1.6 \text{ kg}\) in studies) due to the excretion of excess glucose in the urine.

Metformin enhances the body's sensitivity to insulin, while dapagliflozin operates independently of insulin, allowing for more comprehensive metabolic control.

The combination often results in a mild reduction in systolic blood pressure.

A single fixed-dose combination (FDC) tablet is more convenient for patients than taking separate medications. 

Dosage & Administration

To be taken as directed by the physician, the usual dose is one tablet once daily with meals to minimize gastrointestinal side effects or as prescribed based on patient condition.
